Anatomy of somatic embryogenesis in Carica papaya L. BABT

Mature zygotic embryos of Carica papaya L. ‘Sunrise Solo’ were used as explants for embryogenesis induction. The explants were inoculated on Murashige and Skoog culture medium supplemented with 2 mg.L-1 2,4-dichlorophenoxyacetic acid and incubated in darkness at 25+2°C. Histological analysis of callogenesis and somatic embryogenesis indicated occurrence of direct and indirect somatic embryogenesis development. Direct somatic embryo formation was observed from hypocotyledonary epidermic cells only from explant 18 days after inoculation. Bauhinia forficata link shoot regeneration: histological analysis of organogenesis pathway BABT

Plant regeneration was achieved from cells of callus induced from hypocotyl segments of Bauhinia forficata on half strength Murashige and Skoog culture medium supplemented with several concentrations of BAP. Within 40 days of culture shoot buds formation was observed on callus surface. Calli were then transferred to a same composition culture medium without plant growth regulator in order to induce shoot elongation. Histological studies indicated that in vitro plant regeneration in B. forficata occurred through indirect organogenesis. Meristemoids consisting of small cells with dense cytoplasm and prominent nuclei were randomly distributed throughout the callus surface indicating early stages of shoot bud differentiation. Calcium, potassium and magnesium treatment of Chrysanthemum morifolium cv. "Bi Time" and callogenesis in vitro Scientia Agricola

The chemical composition and vegetative vigor of the donor plant are essential for the satisfactory performance of explants in vitro. In order to test the effect of potassium, calcium and magnesium nutritional status of Chrysanthemum morifolium plants on callogenesis in vitro, pot plants growing in sand were irrigated with nutrient solution containing different levels of potassium (0; 58.5; 117 and 234 mg L-1), calcium (0; 50; 100 e 200 mg L-1) and magnesium (0;12; 48 e 96 mg L-1). Growth of Bauhinia forficata Link, Curcuma zedoaria Roscoe and Phaseolus vulgaris L. cell suspension cultures with carbon sources Scientia Agricola

Cell suspension cultures of Bauhinia forficata Link, Curcuma zedoaria Roscoe and Phaseolus vulgaris L. were used to test plant ability to utilize an alternative to sucrose as carbon source and energy for growth. Glycerol, sorbitol and galactose were the alternative carbon sources tested. Cell suspension cultures established on liquid medium containing sucrose were transferred to culture medium supplemented with sucrose or glycerol, or sorbitol, or galactose as the sole carbon source. Fresh and dry weight increasing and protein content showed marked differences among the different carbon sources used. Sucrose was the best carbon source for all the three plant species tested. Galactose and glycerol promoted slow or no growth of the three studied species....

Histological analysis of the callogenesis and organogenesis from root segments of Curcuma zedoaria Roscoe BABT

Callus was induced from root segments taken from in vitro grown plants of Curcuma zedoaria Roscoe. The explants were cultured on agar-solidified Murashige and Skoog medium supplemented with 13.4muM of alpha-naphthaleneacetic acid and 2.2muM of 6-benzylaminopurine at 25ºC in the dark. Histological analysis revealed that callus was formed from the hypertrophied cortical parenchyma cells of the explant. Some of these cells underwent division while the surrounding cells accumulated starch. Callus was capable of shoot bud regeneration after 70 days when it was transfered to liquid medium of the same composition. After 30 days in liquid medium, buds developed from nodular structures. Micropropagation and callogenesis of Curcuma zedoaria Roscoe Scientia Agricola

Curcuma zedoaria Roscoe (zedoary) is a medicinal properties-bearing Zingiberaceae from which rhizomes are commercially exploited. The objective of this work was to establish an in vitro protocol for micropropagation and callogenesis of Curcuma zedoaria Roscoe as alternative to improve plant production, turning economically feasible the exploitation of its secondary metabolites which present medicinal properties. Micropropagation by using shoot apexes produced by rhizome and from in vitro plants were carried out on Murashige & Skoog medium supplemented with 2.0 mg L-1 benzyl amino purine and 30 g L-1 sucrose. Quantificação da micropropagação de Curcuma zedoaria Roscoe Scientia Agricola

Curcuma zedoaria Roscoe, planta pertencente à família Zingiberaceae que se propaga vegetativamente, tem sido utilizada amplamente por suas propriedades terapêuticas. O interesse na propagação em massa de plantas medicinais in vitro tem aumentado sensivelmente, pois os métodos convencionais de propagação limitam o potencial de utilização destas plantas. Ápices caulinares foram inoculados em meio de cultura sólido contendo os sais básicos e vitaminas de Murashige & Skoog, suplementado com 2,0 mg L-1 de BAP e 30 g L-1 de sacarose. As culturas foram incubadas em sala de crescimento a temperatura de 25±2°C com fotoperíodo de 16/8 horas de claro/escuro. Transferências e repicagens foram realizadas a cada 40 dias. Sucrose metabolizing enzymes in cell suspension cultures of Bauhinia forficata, Curcuma zedoaria and Phaseolus vulgaris PAB

The objective of this work was to study the activity of sucrose metabolizing enzymes in extracts of cell suspension cultures of Bauhinia forficata Link, Curcuma zedoaria Roscoe and Phaseolus vulgaris L. Invertase pathway was identified in the three studied species. Sucrose synthase pathway was also responsible for sucrose metabolism in Curcuma zedoaria and Phaseolus vulgaris cells. Activity values higher than 300 nmol min-1 mg-1 of protein were found for acid and neutral invertases, UDPglucose pyrophosphorylase and phosphoglucomutase in the cell extract of the three plant species. Indução de calogênese eficiente e multiplicação celular rápida são pré-requisitos fundamentais em biotecnologia de plantas. Sucesso na calogênese é dependente dos componentes do meio de cultura e da qualidade dos explantes. Neste trabalho é relatada a influência do tratamento de matrizes de Dianthus caryophyllus L. com nitrogênio na indução de calogênese in vitro. Mudas de cravo cultivadas em vasos contendo areia foram tratadas com soluções nutritivas contendo 5 níveis de nitrogênio.
